Patients should urinate before sperm sampling. The whole ejaculate, which is best brought in a sterile urine sampling bottle, is given for microbiological examination.

Semen culture is usually routinely done with native semen analysis because semen makes up over 99% of prostate secretions and seminal vesicles and less than 1% of sperm. Since the prostate is a reservoir of bacteria in the lower parts of the genital tract, semen culture often provides valuable information about a hidden infection of the lower urogenital tract that is not detected by urine culture or urethral swab.
